When I was an intern in college I saw another intern do this to a senior engineer and it was absolutely classic - it's too good not to share!
Iv'e done this before, it's a classic. Another tip: before you set the taskbar to autohide move it to the top of the screen (or bottom if you target already has it at the top) that way when they are fumbling arround trying to get something to work they are less likely to accidentally make it unhide.
even better than moving all thier icons on a windows machine is to right-click, Arrange Icons By >> uncheck 'show desktop icons'.

I remember first doing this when were in freshmen year of high school and just leaving the computer like that when class ended.
When I worked at a TV station at my university, we did this to an anchor there while she was really stressed out and working on a story but stepped away for a moment. She almost blew up but after we told her it was a joke, she laughed with us and it eased her stress a little.
Another great thing to do is create a crapload of folders, sceenshot that, delete the folders and set it as the background and watch as they try and delete them :D
Did you only get a computer this year. That prank is as old as the ENIAC.
What a waste of time...
Even better than hiding the taskbar, kill the explorer.exe process. That way, all that's left will be the desktop image. This also avoids the need to physically move all the icons off the desktop, because the desktop icons and start menu won't be there.

Just throw their computer in the toilet. Its a great gag. The looks on your co-workers face is priceless. Then when they have that crazy look on their face after seeing their computer in the toilet, have another co-worker sneak up behind them and slip a burlap bag over their head. Then promptly kick them in the crotch. You and your other co-workers will laugh all morning!

One time, back in college I threw my roommates computer in the bathtub while he was taking a bath. He fried up like an egg. I loved it. Really stinky though. Then I took his corpse and ground it up and then put him in a cake which i sent to his mother. It was a great practical joke. I still chuckle about it to this day.

Anyhow. Another variant is to create a single folder of somthing they'd rather not want. Horse porn for example. Take screenshot, delete folder. FUN.
