Primers!! For my first few months in my grad lab, none of my reverse primers were the reverse complement of the sequence--just the reverse. And I wondered why my PCR didn't work! And then, for awhile, I was ordering primers with faulty restriction sites on the ends of them. *sigh* I swear I ordered the same one the wrong way three times. Poor Advisor.....
The way to get really depressed is to think about the overhead that goes to the school. Here at LargeU, it's 52%, so 52% of the grant total goes to back to the school. You know, to run our buildings...and the history buildings...etc etc.
